CRTR1, a novel developmentally regulated transcriptional repressor

Description
CRTR1 was identified as a cDNA expressed specifically in the pluripotent cells of the preimplantation embryo and down regulated at about 4.75 d.p.c. Secondary sites of expression were detected in the embryonic and adult kidney where transcripts were restricted to the epithelial monolayer lining the distal convoluted tubules. Isolation of full length cDNAs revealed that the protein encoded a novel member of the CP2 family of eukaryotic transcription factors. In common with other members of the family, in vitro translated CRTR1 has been shown to bind specifically to the consensus CP2 family DNA binding sequence, and to form hetero-oligomers with other members of the family (collaboration with Steven Jane, Melbourne). In contrast to other CP2 family members, the CRTR1 protein does not appear to direct transcriptional activation but instead represses transcription, apparently when located at the promoter. The repressor domain was localised to the N-terminal 52 amino acids of the protein. The developmental regulation and repressive effects of CRTR1 distinguish this protein from all other members of the family. A transcriptional screen of CP2 family members revealed a number of splicing variants, altered at regions of potential functional significance. The biochemical effects of these alterations are being assessed by in vitro analysis.
