What causes ankylosing spondylitis and who is at risk? Less than 1% of the UK population suffer from the condition. Ankylosing spondylitis is more common and more severe in men than in women. It usually begins between the ages of 16 and 40 years. Although the cause of the condition is unknown, a very strong association is found between ankylosing spondylitis and people who have a blood type called 'HLA-B27'. It is thought that this blood type causes the body's immune system to attack the joints of the spine. Therefore, ankylosing spondylitis is often thought of as an 'autoimmune disease' - a disease in which the immune system attacks the body. Objective of the Autoimmunity Group
Our overall objective is to investigate the initiation of autoimmune diseases and to identify predisposing features of the immune response. Our long-term goal is to define novel vaccine strategies for the prevention of autoimmune disease. In addition we hope to identify factors that may lead to autoimmune disease when using conventional vaccine strategies, so that these can be avoided.
Autoimmune conditions are among the most chronic and debilitating diseases that affect the developed world. Immunological studies of these conditions in man, are largely restricted to investigation of the immune responses in these individuals after the onset of disease has occurred. Because of the lag time between onset and diagnosis, this may be weeks, months, or even years after the initial events that triggered the disease process.
The use of transgenic (TG) mice allows us to work both with known antigens, where the target self-antigens of disease are currently unknown, as well as to express human genes which have been shown by epidemiology to be important in the development of disease. Our approach is to use two systems; a novel antigen-based system to study reactive arthritis (ReA) in an animal model and the well-established model, experimental allergic encephalomyelitis (EAE).

It is characterized by pain, swelling, stiffness, deformity, and/or a diminished range of motion. More than 50 million Americans suffer from osteoarthritis, rheumatoid arthritis, and related conditions, such as fibromyalgia, gout, lupus, Lyme disease, psoriatic arthritis. , joints. Joints of the body are found at the knees, wrists, elbows, fingers, toes, hips, and shoulders. The neck and the back also have joints between the bones of the spine. There are six different types of synovial joints (hinge, ball-and-socket, and so on), but although the types of motion they allow are different, their underlying physiological structure is essentially the same. Two or more adjoining movable bones, whose adjacent surfaces are covered with a layer of cartilage, are surrounded by a fluid-filled capsule made up of ligaments (tough, fibrous tissue). The third is secreted by a thin membrane, the synovial membrane, that lines the inside of the joint capsule. This viscous fluid along with the smooth, rubbery, blue-white cartilage that covers the ends of the bones normally work together to allow the bones within the join to glide smoothly past one another.

Martin Stocks, Ph.D The human immune system is undoubtedly one of the masterpieces of evolution. The processes of gene rearrangement and somatic hypermutation allow for the generation of an immense range of binding specificities in both antibodies and T-cell receptors. Almost any foreign material which find its way into the human body can be targeted and removed or destroyed with exquisite specificity. However, the immune system has not arisen in isolation but is the result of a biological arms race, conducted over millions of years, between man and the microorganisms that can enter the body and cause disease. As the immune system has developed, microorganisms have evolved strategies to subvert or evade its surveillance.
For the microbes, one logical tactic in this battle has been the development of camouflage. If the immune system cannot distinguish an invading microbe from its own tissues, then the battle is all but won. It is therefore logical that microbial or viral pathogens should evolve surface antigens that share sequence homology with normal cellular proteins, or at least present a similar surface structure to the immune surveillance mechanisms.
Since this hypothesis was first proposed in the early eighties, a huge body of evidence has accumulated supporting the view that a number of diseases are caused by molecular mimicry. Over the next several months, this column will review autoimmune disorders. The coverage of this topic is due to the recent resurgence of interest among researchers, clinicians, and families in autoimmunity hypotheses and their possible involvement in the etiologies of childhood neuropsychiatric disorders.
