Als kind wijdde hij zich reeds aan, wat hij noemde, "het ongekunstelde beroep van musicus". Dowlands carrière als luitist begon in Frankrijk. Hij verbleef van 1579 tot 1584 te Parijs in dienst van Sir Henry Cobham, de Engelse ambassadeur in Frankrijk. Daar ontmoet hij luitvirtuozen van die tijd zoals Edinton, Vaumesnil, Perrichon en Bocquet. Gedurende deze periode bekeert hij zich tot het katholicisme. In 1588 wordt hij Bachelor of Music van de universiteit te Oxford. Zijn pogingen om in de Chapel Royal te worden opgenomen waren, vanwege zijn godsdienst, tevergeefs. Hij vertrekt dan ook naar het continent. Eerst naar Duitsland waar hij was uitgenodigd door de hertog van Braunschweich. Hij krijgt er in 1595 een "excellent masters" diploma en maakt kennis met de luitist Gregorio Howet van Antwerpen, zanger en instrumentalist Alessandro Orologio en Johann Cellarius. Na Duitsland komt Italië aan de beurt. In Venetië ontmoet hij de koorleider van de San Marco basiliek: Giovanni Croce. Van Venetië reist hij naar Firenze waar hij voor de hertog optreedt. Van daaruit gaat het naar Rome 'om te studeren' met de beroemde madrigalist Luca Marenzio.

He held musical posts in Paris and Germany, and from 1598 to 1606 he was lutenist to Christian IV of Denmark. In 1612 he obtained a long-desired appointment in England as one of the king's lutenists, a post he held until his death. His lute songs display sensitive text declamation; the accompaniments are subordinate, yet possess some melodic independence. His First Book of Songs or Ayres (1597) was the most often reprinted music book of its time.
